چکیده

BACKGROUND Degenerative diseases are a medical, social, and economic problem worldwide. The most significant factors predisposing the development of degenerative changes in intervertebral discs are a low density and poor biosynthetic potential of the cells. Therefore, stem cell therapy in this case should show high clinical efficiency. METHODS The research aim was to evaluate the regenerative potential of cryopreserved mesenchymal stem cells (MSCs) upon degenerative changes in intervertebral discs. Rats with simulated degenerative damage of the intervertebral disc Co6-Co7 were administrated with 0.5 × 10⁶ of either native or cryopreserved cells on a collagen sponge to the defect area. The results of experiments were histomorphometrically evaluated on the 30th, 60th, and 90th days after treatment. RESULTS The restoration of tears, clefts, and collagen fiber fragmentations was noted on the 60th and 90th day after administration of native and cryopreserved MSCs respectively. An increase in fibrochondrocyte density got ahead of the annulus fibrosus height recovery. In the control group without treatment the regeneration was hardly observed. CONCLUSION The use of MSCs promotes the restoration of the degenerated intervertebral disc. Cryopreserved MSCs have a "lag" therapeutic effect at the early stages, but show similar results to the native analogue on the 90th day after administration.

Degenerative Disc Disease: A Review of Cell Technologies and Stem Cell Therapy

Background & Aim: Low back pain is broadly documented as one of the most widespread pathologies in the advanced domain. Although the reasons of low back pain are uncountable, it has been meaningfully related to intervertebral disc degeneration. Present therapies for Intervertebral Disc (IVD) degeneration such as physical therapy and spinal fusion reduce symptoms' severity, but do not treat the ...

Regeneration potential and mechanism of bone marrow mesenchymal stem cell transplantation for treating intervertebral disc degeneration.

Intervertebral disc degeneration is a primary cause of low back pain and has a high societal cost. The pathological mechanism by which the intervertebral disc degenerates is largely unknown. Cell-based therapy especially using bone marrow mesenchymal stem cells as seeds for transplantation, although still in its infancy, is proving to be a promising, realistic approach to intervertebral disc re...

Mesenchymal Stem Cell in the Intervertebral Disc

Degeneration of the intervertebral disc (IVD) is a major spinal disorder that causes back pain. Nucleus pulposus (NP) in the central of IVD dehydrates and become more fibrous in the IVD degeneration. NP cells undergo apoptosis with the degeneration of extracellular matrix (ECM) components. To replenish the NP cells and core ECM, bone marrow mesenchymal stromal cells (BMSCs) have been highlighte...

Effects of implantation of bone marrow mesenchymal stem cells, disc distraction and combined therapy on reversing degeneration of the intervertebral disc.

Although success has been achieved with implantation of bone marrow mesenchymal stem cells (bMSCs) in degenerative discs, its full potential may not be achieved if the harsh environment of the degenerative disc remains. Axial distraction has been shown to increase hydration and nutrition. Combining both therapies may have a synergistic effect in reversing degenerative disc disease. In order to ...

Thermally Triggered Injectable Hydrogel Which Induces Mesenchymal Stem Cell Differentiation To Promote Regeneration Of The Intervertebral Disc

Introduction: Instability of the motion segment as a result of intervertebral disc (IVD) degeneration is well known as a major cause of low back pain (Balague et al 2011). Degeneration in the central nucleus pulposus (NP) is characterised by a loss of extracellular matrix (ECM) components including proteoglycans particularly aggrecan, and a switch in collagen synthesis from collagen type II to ...
